Plant growth promoting bacteria can improve growth and yield of agronomic plants through direct and indirect mechanisms. Minimizing consequences of using high amount fertilizers and obtaining sustained level is the aims sustainable agriculture. Using of microorganisms due to improving soil fertility and nutrients uptake, lead to increasing growth and yield of plants and ameliorate soil health. The aim of this research was to isolation and purification of bacterial isolates from animal manures, evaluation of their plant growth promotion characteristics and identification of selected isolates.Material and methods: In order to characterization of growth promotion characteristics of bacteria in this study, 10 samples of animal manures from different regions were collected. 27 isolates based on halo production, growth rate and morphological characteristics in NBRIP medium were purified and then and their typical PGP including auxin, Siderophore, HCN production, insoluble phosphates solubilization and secretion of protease, Cellulase, α-amylase and phytase were evaluated and finally selected isolates based on 16SrRNA were identified.Results: Among all of the isolates, maximum phosphorous from tri-calcium phosphate (373 mg/l) and aluminum phosphate (13.3 mg/l) was released by isolates NO. 7. Significant negative correlation (r= -0.86, P≤ 0.01)between maximum solubilization of tri-calcium phosphate and pH was observed. Isolate NO.21 produced greatest amount of auxin (52.8 mg/l). Only isolates NO. 30 and 53 were positive in HCN production. Among 27 isolates only three of them had the ability to secret protease, cellulase and α-amylase. The utmost halo was measured by isolate NO.49 in production of these three enzymes. Evaluation of extracellular phytase production showed that only 6 isolates capable to produce the enzyme. The analysis of the 16S rRNA of the selected isolates showed highly similarity to bacteria belonging to the genera Enterobacter, Sphingobium, Serratia. Only one of the three selected isolates produced weak halo in CAS-agar medium and another two isolates didn't show the siderophore prodction capability.Conclusion: Plant growth promotion characteristics of bacterial isolates, in this research, showed that some of the isolates had the potential to use as PGPB. Those isolates capable of producing protease, amylase and cellulase enzymes not only have usability in agriculture but also because of the importance of this group of enzymes, especially protease and amylase in industry, can also be considered.Keywords: tri-calcium phosphate, aluminum phosphate, auxin, phytase, plant growth promotion}, keywords = {tri-calcium phosphate,aluminum phosphate,Auxin,Phytase,plant growth promotion}, title_fa = {بررسی برخی ویژگی های افزاینده رشدی جدایه های باکتریایی بدست آمده از کود‌های جانوری و شناسایی جدایه‌های برگزیده}, abstract_fa = {سابقه و هدف: رشد گیاهان در خاکهای کشاورزی وابسته به فاکتورهای زیستی بسیاری می‌باشد. ﺑﺎﮐﺘﺮیﻫﺎی افزاینده رﺷﺪ گیاه از راه های ﻣﺴﺘﻘﯿﻢ و ﻏﯿﺮ ﻣﺴﺘﻘﯿﻢ ﻣﯽﺗﻮاﻧند مایه اﻓﺰاﯾﺶ رﺷﺪ و کارکرد در ﮔﯿﺎﻫﺎن زراﻋﯽ ﺷﻮد. کاهش پیامد های ناخواسته بهره گیری بیش از اندازه نهاده‌های کودی و رسیدن به سطح پایدار و مطلوب تولید از اهداف کشاورزی پایدار است. بهره گیری از ریزجانداران به دلیل بهبود حاصلخیزی خاک و جذب عناصر غذایی، منجر به افزایش رشد و کارکرد گیاهان و بهبود سلامت خاک می شود. هدف از این پژوهش جداسازی و ناب سازی باکتری‌ها از کودهای جانوری، ارزیابی ویژگی‌های افزاینده رشدی گیاه آن‌ها و شناسایی جدایه‌های برگزیده بود.مواد‌و روش‌ها: در این پژوهش برای شناسایی ویژگی های افزاینده رشد باکتری‌ها، از ده نمونه کود جانوری سرزمین های گوناگون، 27جدایه در محیط NBRIP جامد بر پایه پیدایش هاله، تندی رشد وویژگی های مورفولوژیک، ناب‌سازی شدند و سپس از دیدگاه ویژگی های PGP شامل توان ساخت اکسین، سیدروفور، HCN، حل کردن فسفات های نامحلول وتراوش آنزیم های پروتئاز، سلولاز، آلفا آمیلاز و فیتاز ارزیابی شد و در پایان جدایه‌های برگزیده بر پایه 16S rRNA شناسایی شدند.یافته‌ها: از میان جدایه‌های بررسی شده جدایه‌ی شماره 7 با آزادسازی 3/13و373 میلی‌گرم در لیتر فسفراز فسفات آلومینیوم و تری کلسیم فسفات بیشترین توان انحلال فسفات‌های نامحلول را نشان دادند. همبستگی منفی معنی‌داری (, p≤0.0186/0-=r) میان توان انحلال تری‌کلسیم فسفات و pH وجود داشت. همچنین جدایه 21 بیشترین میزان اکسین را به اندازه 8/52 میلی‌گرم در لیتر فراوری نمود. تنها جدایه‌های 53 و30 توان ساخت HCN را دارا بودند. از مجموع 27 جدایه باکتریایی بررسی شده تنها سه جدایه توان تراوش هر سه آنزیم پروتئاز، آلفا آمیلاز و کربوکسی متیل سلولاز را فرآوری نموده و جدایه 49 بیشترین هاله را در اندازه‌گیری هر سه آنزیم نشان داد. سنجش فعالیت فیتاز برون یاخته ای نشان داد که تنها 6 جدایه توان ساخت این آنزیم هستند. آنالیز توالی‌های 16S rRNA جدایه های برگزیده نشان داد که جدایه های یاد شده همانندی بسیاری به باکتری‌های جنس های Enterobacter, Sphingobium, Serratia داشتند. تنها یکی از سه جدایه برگزیده هاله اندکی در محیط CAS-agar پدید آورد و دو جدایه دیگر بدون توان ساخت سیدروفور بودند.نتیجه‌گیری: ویژگی‌های افزاینده رشدی گیاه در جدایه‌های باکتریایی بررسی شده در این پژوهش نشان داد که برخی از جدایه‌ها توانایی بهره گیری بالقوه به عنوان باکتری افزاینده رشد گیاه را دارا می‌باشند. آن دسته از جدایه‌های توانمند در ساخت آنزیم های پروتئاز، آمیلاز و سلولاز نه تنها دارای توانایی بهره گیری در بخش کشاورزی هستند بلکه وابسته به اهمیت این دسته از آنزیم ها به ویژه پروتئاز و آمیلاز در صنعت می‌توانند از این نظر نیز مورد بررسی قرار گیرند.}, keywords_fa = {تری‌کلسیم فسفات,فسفات آلومینیوم,اکسین,فیتاز,افزاینده رشد گیاه}, url = {https://ejsms.gau.ac.ir/article_2771.html}, eprint = {https://ejsms.gau.ac.ir/article_2771_ab3b8d409cc40994513612b98d11cfd0.pdf} }
